DL-2-Pyrrolidone-5-Carboxylic Acid - Study on the combined toxicity of quorum sensing inhibitor, sulfamethoxazole and doxycycline hydrochloride to Escherichia coli and its mechanism

Abstract:
quorum sensing inhibitor (QSIs) has the characteristics of no resistance, which is used as a possible alternative to antibiotics, has broad application prospects, therefore, there is the possibility of combined exposure with traditional antibiotic environment, but there is still a lack of related joint effects. In this paper, Escherichia coli (Escherichia coli) as the test organism, seven QSIs(DL-pyroglutamic acid ,N-vinylpyrrolidone, furanone acetate, 2-methyltetrahydrofuran-3-one, 3, 4-dibromo-2 (5h)-furanone,(R) the combined toxicity of 3-pyrrolidinol and D-prolinol with sulfamethoxazole (SMX) and doxycycline hydrochloride (DH) was investigated. According to the analysis of the results, the first 5 QSIs act on the quorum sensing system mediated by AI-2 signaling molecules, and compete with the AI-2 signaling molecules to bind Lsr B protein, the action pathway of DH does not affect each other, so the combined effect is additive; The latter two QSIs act on the quorum sensing system mediated by AI-1 class signaling molecules, and compete with AI-1 class signaling molecules to bind to Sdi protein A, however, the effects of SMX and DH may stimulate the expression of Sdi A protein, so that more QSIs should be consumed to bind to Sdi A, so the combined effect is antagonistic. This study can provide a theoretical basis for the ecological risk assessment of the combined exposure of traditional antibiotics and QSIs.
